After finishing second in Saturday's sprint to team-mate Lando Norris, the pair swapped places in the race as McLaren enjoyed a perfect weekend in the United States.
World champion Max Verstappen started on pole, but was unable to compete with the power of the two McLarens, and dropped out of the top three.
Verstappen and Norris renewed their title battle from last season on the first lap, with the Dutchman locking up heading into the third corner as the McLaren looked to pounce.
However, Norris lost four positions from second after he ran wide, but he was able to quickly pass the Mercedes duo of Kimi Antonelli and George Russell.
Contact between Jack Doohan and Liam Lawson at Turn 1 brought out the virtual safety car to remove debris and, after the lap 4 restart, McLaren asserted their dominance.
Piastri started hounding Verstappen for the lead soon enough and got the job done into Turn 1 on lap 14, with the Red Bull driver locking up as he went into the corner.
Their battle had allowed Norris to close up to them, after wasting little time passing the drivers between him and the leaders.
The Briton initially passed Verstappen in Turn 7, but forced both cars off the track, handing the position back before getting the job done on the following lap.
Verstappen quickly dropped back from the McLarens, losing nine seconds in as many laps before his pit stop, while a virtual safety car was deployed during his pit-stop.
That allowed Russell, who was the only driver in the top 10 to start the race on hard tyres, to pit for fresh tyres and re-join still ahead of Verstappen.
From the lap 30 restart onwards, Piastri comfortably protected his lead against Norris, who shaved off a few seconds, but never looked like closing the gap to his team-mate.
Russell was able to claim a fourth podium of the season by finishing third, while Verstappen finished one place further back.
Alex Albon secured another huge points haul for Williams by coming in fifth, ahead of Kimi Antonelli, while Ferrari endured another difficult day on the track.
Lewis Hamilton settled for eighth and took several heated shots at his Ferrari engineer over his frustration at a delayed swap with Charles Leclerc, who ended up in seventh.
Carlos Sainz and Yuki Tsunoda rounded off the top 10, with Doohan, Lawson, Gabriel Bortoleto and Oliver Bearman all failing to finish.

The Australian has also now finished 34 consecutive races in the competition, equalling Daniel Ricciardo between Styria 2020 and Mexico City 2021 as the third-longest run in F1 history.
It was also a fine weekend for McLaren, who extended their lead in the constructors' championship. They have now won five of the first six races, equalling their best ever start to a season, which came back in 1998.
