The Spaniard, who was making his return to the tour after missing the Madrid Open due to an adductor injury, was always in control of the encounter and won 6-3 6-3 in an hour and 22 minutes.
The four-time grand slam champion did have his knee wrapped but seemed to be moving freely as he aims for his first-ever Italian Open title.
Alcaraz made a fast start, winning the first four games, though he was broken when serving for the set at 5-2.
He subsequently broke the Lajovic serve straight away to take the first set, however.
The Spaniard went a break up again early in the second set and managed to maintain his level to wrap up a comfortable victory.
The third seed is vying for his third title of the year and his second Masters 1000 crown of the campaign, having already triumphed in Monte-Carlo.
Alcaraz will face either Estoril Open winner Alex Michelsen or Laslo Djere in the next round.
Data Debrief: Clay dominance continues for Alcaraz
Alcaraz has already had great success on clay in his young career, winning the French Open in 2024 and another eight titles on the surface on the ATP Tour.
His winning percentage on clay stands at 82.7%, trailing only Rafael Nadal (90.5%) since the inaugural ATP Tour season in 1990.
Alcaraz also extended his 100% record against Lajovic, with the head-to-head now at 5-0 in the Spaniard's favour.
