Sinner needed just 90 minutes to earn a 6-4 6-2 victory as De Jong struggled with an injury sustained after a fall in the second set.
The world number one forced two break points in the opening game with no reward before going on a three-game winning streak to take control.
However, De Jong wrestled back some control with back-to-back breaks of his own before losing on serve again, allowing Sinner to take the first set.
Sinner again got an early break in the second, though De Jong kept the pressure on before slipping and landing awkwardly on his wrist, with the top seed making his way across the court to help him back to his feet.
Though the Dutchman won that game, after a medical time-out, Sinner won the next three, serving and breaking to love to finish off the match.
He will face Francisco Cerundolo for a place in the quarter-finals.
