The Brit, who earned his maiden Formula One win at the Miami Grand Prix last year, had started third on the grid, but he found an edge over McLaren team-mate Oscar Piastri, who finished second, thanks to the safety car.
Lewis Hamilton, who won the sprint at the Chinese Grand Prix earlier this season, continued his impressive form in the format by finishing third.
After a delayed start due to the weather, pole-sitter Kimi Antonelli, who is the youngest person to take the position in either of F1's qualifying formats, immediately lost out to Oscar Piastri, Lando Norris and Max Verstappen.
A collision between Fernando Alonso and Liam Lawson on lap 15 then caused the safety car to come out, with the timing benefitting Norris.
He pitted one lap after Piastri, who had led since the first corner, and rejoined in the lead, seeing it out to the end.
However, the drama had started before the race had even begun, with heavy rain causing Charles Leclerc to crash into the wall before making it to the grid.
He was unable to compete, leaving him in a race to get the car ready for qualifying later in the day.
There was still more upheaval to come, though. Verstappen made contact with Antonelli in the pit lane, resulting in a 10-second penalty for an unsafe release.
Despite crossing the line in fourth, the Dutchman was demoted to 17th by the penalty. Antonelli, meanwhile, finished outside the points in 10th.
Data Debrief: Norris' luck in Miami keeps going
Piastri was left to rue his frustrating lucklessness in Miami, but it is turning out to be a special place for Norris, who clinched his second sprint win.
His first came in Sao Paulo last year, and he is the third driver to win twice in the format, level with Valtteri Bottas and Piastri.
As for Verstappen, he was not able to add to his 11 sprint victories, having now failed to finish on the podium in the last four such races.
